SMTP2 面向需要自动吸排液与状态反馈的液体处理工位。同一套移液组件在取样、试剂加入和分装时承担不同任务,检测信号也应与各步骤的实际异常对应。
样本前处理:从原始样本管转移到处理容器
在样本前处理模块中,移液单元先装载吸头,寻找液面,再吸取规定体积的样本并移至目标容器。这一任务称为样本转移(sample transfer)或样本移取(sample pipetting),关键是吸液位置、实际吸入量和更换样本时的残留控制。
SMTP2 的液面与吸头检测可用于设备的取样状态判断,尖端堵塞检测可为异常吸液处理提供信号。不同样本液位、黏度及容器底部形状仍需配合运动轴和吸液参数验证。
分析流程:试剂加入与反应体系配制
把试剂加入反应管或板孔属于试剂分配(reagent dispensing);按配方依次加入样本、试剂和缓冲液则属于反应体系配制(reaction setup)。同一移液单元在这两个步骤中,都需要把设定体积完整送到目标容器,并控制样本之间的携带污染(carryover)。
SMTP2 可为此类自动加样工位提供吸排液与检测信号。针对具体液体建立移液参数组合(liquid class),将吸液速度、停留时间、离液动作和排液高度配套设置;一次性吸头的更换时机也应写入同一流程。
样本分装:从一个源容器分配到多个样品管
把同一源液按份转移到多个容器称为分装(aliquoting)。逐管吸排适合分别管理每次转移;一次吸液后多次分配称为多次分液(multi-dispensing),重点是首份、中间份与末份是否达到相同的体积要求。
选用 SMTP2-1000 μL 时,应把计划分配总量与方法所需余量一起核对吸头和泵的可用容量。若采用多次分液,需在设备指令和动作流程中实现,并分别检查首末份体积、转移途中滴液和缺液后的处理。
